Product Description & Scientific Applications
1,3,5-Trimethoxybenzene (phloroglucinol trimethyl ether) is an electron-rich arene. It is a key component of the scent of the Chinese rose.
Quantitative NMR. 1,3,5-Trimethoxybenzene is used as an internal standard in quantitative ¹H NMR (qNMR) spectroscopy. In pharmaceutical analysis, it has been used as a qNMR standard for compliance testing of active substances and approved medicinal products, and for the analysis of suspected illegal medicines. In synthesis, it is added to reaction mixtures as an internal standard so that yields can be determined by ¹H NMR, and it is used to measure the purity of synthesised compounds by qNMR.
Removal of p-methoxybenzyl groups. Combined with a catalytic amount of silver(I) hexafluoroantimonate, 1,3,5-trimethoxybenzene removes the p-methoxybenzyl (PMB) protecting group from alcohols and from an acid. The method is mild and chemoselective.
General reaction: R–O–PMB (alcohol or acid protected as PMB) → R–OH, with catalytic AgSbF₆ and 1,3,5-trimethoxybenzene.
Electrophilic aromatic substitution. With N-fluorobenzenesulfonimide (NFSI) as the electrophile, 1,3,5-trimethoxybenzene gives fluorinated products, and no amidation products are detected.
substitution
General reaction: 1,3,5-trimethoxybenzene + NFSI → fluorinated products (no amidation products detected).
