Product Description & Scientific Applications
5-Methylcytidine (m5C) is a modified ribonucleoside.
- RNA modification. 5-Methylcytidine occurs in tRNA and rRNA in bacteria, archaea and eukaryotes. Transcriptome-wide mapping has also reported sites in mRNA and other non-coding RNAs.
-
RNA methyltransferases. In animals the methyltransferase NSUN2 modifies specific tRNAs. Human DNMT2 methylates cytosine 38 in the anticodon loop of tRNAAsp.NSUN2, DNMT2⟶(in tRNA)
Reaction (in tRNA): cytidine residue → 5-methylcytidine (m5C) residue.
-
Oxidation to 5-hydroxymethylcytidine. Tet enzymes convert 5-methylcytidine to 5-hydroxymethylcytidine in RNA in vitro. 5-Hydroxymethylcytidine is present in RNA of mammalian cells and tissues.Tet enzymes⟶(in RNA, in vitro)
Reaction (in RNA, in vitro): 5-methylcytidine (m5C) → 5-hydroxymethylcytidine.
- Modified mRNA. Replacing cytidine with 5-methylcytidine in mRNA made by in vitro transcription lowers the innate immune response to the RNA and raises protein expression. Modified nucleosides are used in mRNA vaccines for this reason, the approved COVID-19 vaccines using N1-methylpseudouridine.
- Analytical standard. Synthetic 5-methylcytidine is used to prepare calibration solutions for LC-MS/MS quantification of RNA modifications.
