Product Description & Scientific Applications
7-Amino-4-methylcoumarin (AMC) is a fluorogenic compound and the traditionally used leaving group in peptide substrates for protease and peptidase assays. Released AMC is detected by fluorescence, with excitation at 365 nm and emission at 440 nm. In a tryptophanase assay with a serine-AMC carbamate substrate, this fluorescence read-out allowed routine assay of solutions containing 10⁻³ units of enzyme activity.
Protease assays. In bottlebrush polymers with AMC attached through caspase-3-cleavable peptides, enzymatic cleavage releases free AMC and gives a “turn-on” fluorescence signal. GD₄K-AMC measures the activity of the serine protease enteropeptidase through a shift in its emission spectrum on cleavage.
Specificity profiling. Libraries of tripeptide-AMC substrates are used to profile protease cleavage specificity through specificity constants (kcat/Km). This approach defined the substrate preferences of the Mycobacterium tuberculosis ClpP1P2 protease.
Substrate synthesis. Aminoacyl-AMC derivatives are important synthons for the construction of chromogenic and fluorogenic protease substrates.
Histological staining. AMC is a fluorescent substitute for Schiff’s reagent in periodic acid-Schiff (PAS) staining. Under UV excitation, AMC-stained sections show glycogen- and mucus-rich structures, such as colonic goblet cells, in blue fluorescence.
