Product Description & Scientific Applications
Copper(II) trifluoromethanesulfonate (copper(II) triflate, Cu(OTf)₂) is the copper(II) salt of trifluoromethanesulfonic acid. In organic synthesis it is used as a catalyst, either on its own or combined with chiral nitrogen ligands, such as diamines, oxazolines and bis(oxazolines), for enantioselective reactions.
Mannich-type reactions. With a chiral diamine ligand, Cu(OTf)₂ catalyses enantioselective Mannich-type reactions of N-acylimino esters with silyl enol ethers and with alkyl vinyl ethers. The products are N-acylated α-amino acid esters with a carbonyl group in the side chain. The method has been applied to the synthesis of an inhibitor of ceramide trafficking.
Mannich-type reaction of an N-acylimino ester (left) with a silyl enol ether (centre) gives an N-acylated α-amino acid ester (right).
Henry reaction. In the Henry (nitroaldol) reaction, nitromethane adds to an aldehyde to give a β-nitro alcohol. Complexes of Cu(OTf)₂ with chiral oxazoline ligands catalyse the asymmetric Henry reaction of aldehydes with nitromethane.
Henry reaction of an aldehyde with nitromethane gives a β-nitro alcohol.
Friedel–Crafts reactions. With chiral bis(oxazoline) ligands, Cu(OTf)₂ catalyses enantioselective Friedel–Crafts additions of indoles to N-sulfonyl aldimines. The products are optically active 3-indolylmethanamine derivatives.
Addition of trimethylsilyl cyanide. Cu(OTf)₂ catalyses the addition of trimethylsilyl cyanide (TMSCN) to a range of aldehydes and ketones. The products are cyanohydrins protected as their trimethylsilyl ethers.
Addition of trimethylsilyl cyanide to an aldehyde or ketone (left) gives the trimethylsilyl ether of the cyanohydrin (right).
Annulative amination and carbazole synthesis. Cu(OTf)₂ is also used as a catalyst for annulative amination, and for the synthesis of carbazoles by intramolecular oxidative C–N bond formation with a hypervalent iodine reagent.
