Product Description & Scientific Applications
D-Luciferin (D-LH2) is the substrate of firefly luciferase, the light-producing enzyme of the firefly Photinus pyralis. It consists of a 6-hydroxybenzothiazole joined to a thiazoline carboxylic acid, with the (S) configuration at C-4.
The light reaction. Firefly luciferase needs D-luciferin, ATP, oxygen and a metal ion, usually Mg²⁺. In the first step, the enzyme joins D-luciferin to AMP from ATP and releases pyrophosphate. Oxygen then reacts with this luciferyl adenylate; the product loses carbon dioxide and gives oxyluciferin in an excited state, which emits light centred near 560 nm.
Firefly luciferase oxidises D-luciferin (left) to oxyluciferin (right). AMP and pyrophosphate are also formed.
Reporter gene assays. Mammalian cells have no luciferase of their own, so the firefly luciferase gene is used as a reporter of gene regulation. After D-luciferin is added, the light produced is proportional to the amount of enzyme. Luciferase reporters are widely used in cell-based assays and in high-throughput screening.
Bioluminescence imaging. D-Luciferin is given to mice that carry luciferase-expressing cells, to measure luciferase activity in the living animal.
