Product Description & Scientific Applications

Girard's reagent T, 1-(carboxymethyl)trimethylammonium chloride hydrazide, belongs to the Girard reagents, a family of quaternary ammonium hydrazides. It reacts specifically with aldehyde and keto groups.

The resulting hydrazone carries a precharged quaternary ammonium group, which enables its detection by positive-ion electrospray mass spectrometry. In tandem mass spectrometry, Girard T hydrazones undergo facile neutral loss of trimethylamine, a transition used for multiple reaction monitoring.

Applications and Reactions

  • Ecdysteroid analysis: Derivatization of the 6-ketone group allows LC-MS/MS detection of endogenous ecdysteroids in crude, lipid-rich extracts from Drosophila larvae and pupae.
  • Glycan analysis: It labels reducing glycans as stable hydrazones directly on the MALDI target, and MALDI-TOF-MS then shows only [M]+ ions, free of sodium and potassium adducts. The method applies to reducing glycans from milk, N- and O-glycans from glycoproteins and glycans derived from glycosphingolipids, and it suppresses desialylation during MALDI analysis. The non-reducing saccharides sucrose and raffinose and the sugar alcohol sorbitol do not react.
  • DNA damage analysis: It conjugates the 5-formyl group of 5-formyl-2′-deoxyuridine, a major thymidine lesion generated by reactive oxygen species, for its LC-MS/MS quantification in cellular DNA.
