Product Description & Scientific Applications
Coumarin 7 fluoresces from an intramolecular charge-transfer (ICT) excited state, with the 7-diethylamino group as donor and the 3-(2-benzimidazolyl) substituent as acceptor. It emits from a planar ICT state across the solvent range, stabilised by an intramolecular hydrogen bond between the benzimidazole N–H and the lactone carbonyl. Emission is solvatochromic: in water the neutral dye absorbs at 462 nm and emits near 500 nm with a fluorescence quantum yield of 0.15, while protonation moves absorption to 475 nm and emission to 512 nm and lowers the quantum yield to 0.05. In high-polarity protic solvents, beyond a Lippert–Mataga Δf of about 0.31, a nonfluorescent twisted ICT (TICT) state opens a nonradiative channel, so quantum yield and fluorescence lifetime fall below their linear solvent-polarity correlation.
Common Scientific Applications
- Laser dye. Used in ethanolic solution as a gain medium, and as a laser-induced-fluorescence staining fluorophore.
- Environment-reporting fluorophore. The large solvatochromic and host-induced spectral shifts make Coumarin 7 a reporter of local polarity and confinement.
- Host–guest photophysics. Encapsulation in sulfobutylether-β-cyclodextrin suppresses the TICT channel, doubling the aqueous quantum yield from 0.15 to 0.30 and raising photostability.
- Ratiometric sensing. Driving the monomer–aggregate equilibrium with poly(styrene sulfonate) at pH about 3 gives a readout at 650 nm against the 513 nm monomer band.
