Product Description & Scientific Applications

SPDP (N-succinimidyl 3-(2-pyridyldithio)propionate) is a heterobifunctional reagent: its N-hydroxysuccinimide ester reacts with amino groups, and its 2-pyridyl disulfide group reacts with aliphatic thiols. Reaction with a thiol releases pyridine-2-thione, and its absorbance at 343 nm can be measured to follow the reaction.

HONOO

General reaction: R–NH₂ + SPDP → 2-pyridyl disulfide amide + N-hydroxysuccinimide; R–NH₂ = an amino group, for example of a protein or of a solid support.

Thiolation of proteins. SPDP introduces thiol groups into proteins in two steps: acylation of amino groups, then reduction of the protein-bound 2-pyridyl disulfides with dithiothreitol (DTT). The reduction can leave the native disulfide bonds of the protein intact.

SHN

General reaction: 2-pyridyl disulfide amide → thiol + pyridine-2-thione.

Protein–protein conjugates. A protein without free thiols, once modified with SPDP, reacts by thiol–disulfide exchange with a protein that carries a native or introduced thiol, giving a disulfide-linked conjugate. Enzyme–enzyme, enzyme–albumin and enzyme–antibody conjugates have been prepared in this way. Ricin A chain and gelonin have each been coupled to a monoclonal antibody with SPDP through a disulfide linkage. The disulfide bond between the two proteins can be split by reduction or by thiol–disulfide exchange, so the conjugation is reversible.

RNHOSSR′

SHN

General reaction: 2-pyridyl disulfide amide + R′–SH → disulfide-linked conjugate + pyridine-2-thione; R′–SH = a native or introduced thiol of a second molecule.

Peptide–carrier conjugates. SPDP has been used to link a synthetic peptide to an immunogenic carrier protein through the thiol of a cysteine residue.

Liposomes and nanoparticles. A monoclonal antibody, itself modified with SPDP, has been coupled to liposomes that contain SPDP-substituted phosphatidylethanolamine. Amino-functionalised nanoparticles activated with SPDP have been coupled to a protein carrying an introduced thiol.
